Acid Extends Hematoxylin Life: Why do I Need it for Differentiation?

A mild acid solution is important for clearing the nuclei and reducing overall background staining on the slide. When this step is inadequate, nuclei appear overstained and a purple haze will be noted grossly on the slide. Microscopically, mucin will be stained purple and may obscure details necessary for diagnosis.
