Differentiation of the eosin staining occurs in the subsequent water wash and continues as the tissue goes through the dehydration alcohols.
The desired result should yield the following three shades of pink:
- Orange-pink: Red blood cells
- Light pink: Connective tissue
- Dark pink: Muscle fibers
The cytoplasm of the cells will contain varying shades of pink.
