Like singleplex assays, attention must first be given to:
- Sample collection: Assure a representative, validated sample is processed and ample nucleic acids are present.
- Transport: Assure a system is in place to maintain the stability of nucleic acids (e.g., on ice or with RNA-stabilized media when appropriate).
- Storage: If the sample is not processed immediately, storage occurs most commonly at -80°C.
- Sample processing: Adhere to Standard Precautions.
- Nucleic acid extraction: When applicable, use manual or automated extraction.
- Contamination: Avoid contamination by having an appropriate environment that optimizes containment and adheres to post-processing decontamination (e.g., ethanol and bleach).
